Epithelial-mesenchymal crosstalk is vital for balancing expansion and regression of the progenitor cell pool during tissue turnover. In hair growth cycle, little known about mechanisms underlying regulated death follicle that precedes each regeneration re-growth. Here, we discover dermal sheath smooth muscle, which lines all follicles, controls via transforming factor (TGF)-β mechanosignaling. The secretes TGF-β, first stored as an inactive, latent complex in basement membrane extracellular matrix. then contracts to free TGF-β essential signaling progenitors regulation regression. Finally, identify Integrin-alpha-V on surface molecular link force-mediated release. Our study establishes a mechanosignaling niche reduction through mechanical force, signal activation cell-cell communication.
